How the hematocrit test is done
The hematocrit, or packed cell volume, is the fraction of a blood sample occupied by red cells. It is one of the basic tests for anaemia and for the concentration of the blood. In the micro method, a thin capillary tube is filled with blood, sealed at one end and spun at high speed so the red cells pack into a solid column. The length of the red cell column divided by the total length of blood gives the hematocrit, read directly on the reading device.
- Fill a heparinised capillary with capillary blood, or a plain capillary with anticoagulated venous blood, to about three quarters.
- Seal the dry end by pressing it into the sealing wax tray.
- Place the capillaries in the 24-place rotor with the sealed ends outward, balancing opposite positions.
- Close the lids, set the speed and time, and run the centrifuge.
- Read the packed cell volume of each capillary on the reading device.
With 24 places, a whole batch of patient samples or a class set can be run together, and the stainless steel inner bowl is easy to clean if a capillary breaks.
