How colonies are counted
Counting colonies is the last step of the plate count method. A sample is diluted in steps, a known volume is spread or poured onto agar, and after incubation each visible colony is taken to have grown from one viable cell. Multiplying the count by the dilution factor and dividing by the volume plated gives colony-forming units (CFU) per ml or per gram. Plates with roughly 30-300 colonies are normally chosen for counting.
- Place the incubated 110 mm Petri dish on the counter and view it through the x1.7 magnification.
- Mark each colony on the base of the dish with the marking pen as it is counted, so none is counted twice.
- Use count correction to remove a mistaken count, and hold to keep the total on the display.
- Store the result in one of the 125 memory locations before moving to the next plate.
