How the microtome is used
A microtome cuts tissue into slices thin enough for light to pass through, so cells can be studied under a microscope. The specimen is first fixed, dehydrated and embedded in a paraffin wax block, which supports the tissue while it is cut. In a rotary microtome, turning the drive wheel moves the block up and down past a fixed razor; on each turn the ratchet-and-cone feed advances the block by the chosen thickness, so a continuous ribbon of sections builds up on the blade.
Settings of a few microns suit animal tissues and fine histology, while thicker settings up to 50 microns suit plant material and teaching slides. The razor tilt, set against its reference scale, gives the clearance angle that produces clean, uncompressed sections.
- Clamp the trimmed wax block in the object holder and fit the razor, setting its tilt on the reference scale.
- Set the section thickness at the rear between 1 and 50 microns and check it on the front indicator.
- Turn the drive wheel steadily to cut a ribbon of sections.
- Float the ribbon on warm water, pick up sections on glass slides, then dry and stain them.
- Close the hinged cover over the mechanism and knife holder after use.
