How the TLC experiment is done
In thin-layer chromatography the silica gel layer is the stationary phase and a solvent, the mobile phase, rises up the plate by capillary action. Each component of a spotted mixture is carried up at its own rate, depending on how strongly it is adsorbed on the polar silica compared with how well it dissolves in the solvent, so the mixture separates into a line of spots.
Each spot is described by its retention factor, Rf = distance moved by the spot / distance moved by the solvent front. Comparing Rf values with those of known standards run on the same plate identifies the components.
- Cut a strip from the sheet with scissors and draw a pencil start line about 1 cm from the bottom.
- Spot the sample and any reference substances on the line with a fine capillary.
- Stand the strip in a covered tank with the solvent below the start line.
- Remove the strip when the solvent is near the top, mark the solvent front and let it dry.
- Locate the spots and calculate the Rf value of each.
